CORE DEFINITION
Long-term depression (LTD) is a persistent decrease in synaptic strength lasting from hours to days. It can be induced by strong synaptic stimulation (e.g., in cerebellar Purkinje cells) or by prolonged weak stimulation (e.g., in the hippocampus). LTD is thought to be caused by changes in postsynaptic receptor density, but presynaptic changes may also play a role. Cerebellar LTD is hypothesized to be important for motor learning, and hippocampal LTD may be important for erasing past memories. Hippocampal/cortical LTD can be controlled by NMDA receptors, metabotropic glutamate receptors (mGluR), or endocannabinoids.
